OGG1 activator / An 8-oxo guanine DNA glycosylase 1 (OGG1) activator. Oxidative DNA damage is recognized by OGG1 which then excises 8-oxoguanine, leaving a substrate for apurinic endonuclease 1 (APE1) and initiating DNA repair. TH10785 binds to Phe319 and Gly42 of OGG1 increasing it catalytic activity 10-fold and generating a previously undescribed β,δ-lyase enzymatic function. TH10785 increases OGG1 recruitment to and repair of oxidative DNA damage. Increased repair of oxidative DNA damage by small molecules may have therapeutic applications in various diseases and aging.1
