Action Potential Dose Response
Electrophysiology assay to evaluate compound effects on adult human cardiac action potentials. The assay is performed on ventricular-like iCell® Cardiomyocytes or Cor.4U® Cardiomyocytes. A minimum of four cells are tested per compound. Further evaluation of active compounds on individual ion channels is available.

A. Action potential recording from a single adult human iPSC cardiomyocyte exposed to increasing concentrations of the hERG blocker E-4031. E-4031 significantly prolonged the action potential in a dose-dependent manner. B. Action potential recording of a single adult human iPSC cardiomyocyte exposed to increasing concentrations of verapamil. The lowest dose, 0.1 μM, caused a significant shortening of the action potential due to blockade of the L-type calcium channel; but at a higher dose, 3.0 μM verapamil, the shortening of the action potential duration was reversed due to blockade of the hERG channel. |
